چکیده

The results of lymphocyte functional studies performed with the peripheral blood mononuclear cells (MNCs) of patient Wa with a non-Sezary T-ceIl leukemia are described. The aggressive clinical course and the distinctive histologic, cytochemical, and cytogenetic features of this T-cell lymphoproliferative disorder are detailed in an accompanying report. Immunologic marker analysis revealed that 76% of the Wa MNC fraction formed rosettes with sheep erythrocytes (E-rosettes), and on histologic examination, 95% of the E-rosettes contained neoplastic lymphocytes; 10% of the MNCs formed EAC rosettes; and 1 3% showed SIg positivity by immunofluorescence. In vitro mitogenic responses of Wa cells measured by 3H-thymidine incorporation were extremely low. and responses to candida and streptococcal antigens were absent. Although a normal percentage of B cells was evident in the Wa MNC fraction, no Ig synthesis occurred in either pokeweed mitogen (PWM) stimulated or unstimulated cultures. In coculture experiments with control MNCs from two normal individuals, the Wa cells exhibited no suppressor or helper effects on the normal PWMstimulated lg synthesis. Importantly however, the Wa cells demonstrated suppressor effects on the normal mixed lymphocyte reaction (MLR) occurring between populations of allogeneic MNCs from two normal donors. The degree of MIR suppression increased with the presence of

In Vivo Effects of Calcitriol on Phenotypic and Functional Properties of Dendritic Cells

Background: Dendritic cells (DCs) play a central role in the initiation and expansion of T cell mediated immune responses with potential immunotherapy application. The compounds which have the ability to induce immunomodulatory effects on DCs may be employed for the treatment of immunopathologic conditions such as autoimmune diseases. Objective: The aim of this study was to investigate the in v...

An In Vitro Assay to Evaluate the Immunomodulatory Effects of Unrestricted Somatic Stem Cells

Background: Unrestricted somatic stem cells (USSC) are cord blood stem cells that have been considered as candidates for the regulation of immune responses. Therefore, potential exists for their use in the suppression of immune response after transplantation surgery. Objective: The aim of this study was evaluation of the effect of USSC on mixed lymphocyte reaction (MLR) as a model for graft rej...

Monoclonal antibodies to guinea pig Ia antigens. II. Effect on alloantigen-, antigen-, and mitogen-induced T lymphocyte proliferation in vitro

Four xenographic monoclonal antibodies to guinea pig Ia antigens were tested for their inhibitory effects on antigen-, alloantigen-, and mitogen-induced T cell proliferation. All four monoclonal antibodies reacted with strain 2 Ia antigens, and all four were capable of inhibiting the strain 13 against strain 2 mixed leukocyte reaction (MLR) by 50-70%; the two monoclonals that reacted with strai...

Monoclonal antibody analysis of human T lymphocyte subpopulations exhibiting autologous mixed lymphocyte reaction.

In autologous mixed lymphocyte reaction (MLR), T cells proliferate in response to the stimulation by autologous non-T cells. In the present study, human T cell subpopulations, defined by murine monoclonal antibodies OKT4, OKT8, or 9.3, were examined for their capacity to proliferate in autologous and allogeneic MLR. It was observed that the treatment of responder T cells with OKT4 or 9.3 antibo...
